泡桐丛枝病发生特异相关蛋白质亚细胞定位及质谱鉴定

Subcellular Localization and Mass Spectrum Identification of the Protein Related to Paulownia Witches' Broom Phytoplasma Infection

摘要:

借助胶体金免疫电镜和质谱分析技术,利用制备的专化性抗体,进行豫杂一号泡桐丛枝病发生特异相关蛋白质(m24 ku, pI 6.8)叶片和茎尖的亚细胞定位和质谱鉴定研究。结果显示: 豫杂一号泡桐丛枝病发生特异相关蛋质分布于健康苗叶片和茎尖细胞的细胞壁、液泡和叶绿体部位,患病幼苗叶片和茎尖细胞相同亚细胞器部位未发现该蛋白质的存在,进一步表明植原体侵染阻断了此蛋白的合成。此外,基质辅助激光解吸飞行时间质谱仪(MALDI-TOF-MS)分析该特异蛋白后,通过查询MSDB数据库初步断定该特异相关蛋白为水稻叶绿体分子伴侣Q6K822_ORYSA的同源蛋白。

Abstract:

The subcellular localization and mass spectrum identification of the protein (m 24 ku, pI 6.8) related to witches' broom of Paulownia tomentosa×P. fortunei were investigated with prepared specificity antibody with the colloidal goldlabeling electron microscopy and mass spectrometry techniques. The results indicated that the specific protein was mainly localized in the cell wall, vacuole and chloroplast regions of the healthy seedling leaves and stem apexes cells, while the protein was not found in the same regions of the cell of the infected seedlings, which suggested that the phytoplasma infected interrupted the protein synthesis. Moreover, the protein was preliminarily assessed as the cognate protein of the rice chloroplast molecular chaperone Q6K822_ORYSA after analyzed by matrixassisted desorption ionization timeof flight mass spectrometry (MALDI-TOF-MS) through analysis of the MALDI-TOF-MS data using the public database MSDBI.
