Abstract:
A RAPD marker specific to leaf shape of fig (Ficus carica) was developed and converted into SCAR marker for germplasm identification. RAPD amplifications with random primers S-2082 produced a unique fragment linking to the noncordate base leaf shape. The unique fragment was extracted, cloned and sequenced. Sequencing results indicated that the size of the fragment was 1 640 bp. Specific primers were designed based on its sequence and used in SCAR-PCR, and it was showed that the RAPD marker from S2082-1640 was successfully converted into SCAR marker.
