STUDY ON GENETIC DIVERSITY OF CUNNINGHAMIA LANCEOLATA AND TAIWANIA FLOUSIANA BY USING CHLOROPLAST MICROSATELLITES

Abstract: DNA was isolated from germinating embryos of 5 and 7 natural populations of Cunninghamia lanceolata and Taiwania flousiana, respectively. Two different techniques were used for the DNA isolation. Similar results in terms of quantity and quality of DNA were obtained and the economical one was selected as main method. The best DNA amplification condition was obtained after experiments. AMOVA was studied by using the software of Arlequin and PHYLIP. Results showed that the cpSSR was sensitive in revealing the genetic variation between 2 tree species and among the populations inside species, it is of special importance in studing the evolution history of coniferous tree species, gene resources conservation strategy and tree breeding strategy.
